Effect of anaerobiosis on staphylococcal nuclease production.

چکیده
Five strains of Staphylococcus aureus were examined quantitatively for the production of nuclease under aerobic and anaerobic conditions. Hydrolysis of deoxyribonucleic acid and ribonucleic acid was detected by measuring the release of acidsoluble nucleotides spectrophotometrically. We found that the enzyme was produced anaerobically, as well as aerobically, and that anaerobiosis had no effect on production of this enzyme when other conditions, such as pH, were held constant.

Fermentation studies of the secretion of Serratia marcescens nuclease by Escherichia coli.
The secretion of a Serratia marcescens nuclease was followed by fermentation with Escherichia coli. A plasmid, p403-SD2, carrying a 1.3-kilobase-pair insert with a 0.4-kilobase-pair region upstream of the nuclease gene caused a growth-phase-regulated expression of nuclease in E. coli in the same way as that seen in S. marcescens. Deletion of the regulatory gene generating plasmid p403-Rsa1 resu...
متن کاملComplementation of the Lactococcus lactis secretion machinery with Bacillus subtilis SecDF improves secretion of staphylococcal nuclease.
Unlike Bacillus subtilis and Escherichia coli, the gram-positive lactic acid bacterium Lactococcus lactis does not possess the SecDF protein, a component of the secretion (Sec) machinery involved in late secretion stages and required for the high-capacity protein secretion in B. subtilis. In this study, we complemented the L. lactis Sec machinery with SecDF from B. subtilis and evaluated the ef...
متن کاملStaphylococcal enterotoxin B and nuclease production under controlled dissolved oxygen conditions.
Enterotoxin B, nuclease, and total exoprotein production by Staphylococcus aureus strain S-6 was studied in a 0.5-liter fermentor system. While these extracellular products were elaborated over a wide range of aeration rates, maximal production occurred within the very narrow range of 125 to 150 cm(3) of air per min. The levels attained at the optimal aeration rate were not increased by maintai...
متن کاملAnti-microbial effect of Nigella sativa seed extract against staphylococcal skin Infection
Background : The development of microbial resistance to the existing anti-microbial agents has become a real challenge and a serious problem facing patients suffering from skin infections. Seeds of Nigella sativa have been used for a long time in folk medicine for the treatment of skin infections. Production of new potent agents is urgently needed, especially for hospitals and health care cen...
متن کاملLinked Control of Antinuclease Specificities
Staphylococcal nuclease is a convenient model antigen for study of the genetics of the antibody response partly because of the large amount of information available about its chemistry and because of its relative simplicity among natural protein antigens. However, we have shown in the accompanying paper (1) that even for nuclease the interpretation of the genetic control of the immune response ...
